Метод центрифугирования в градиенте плотности (density gradient centrifugation), в котором в качестве рабочего используется раствор хлористого цезия (получены близкие результаты и для сульфата цезия), градиент плотности образуется и стабильно поддерживается во время центрифугирования при достаточно высокой скорости вращения; молекулы ДНК или другие макромолекулы при центрифугировании достигают определенного слоя раствора хлорида цезия, плотность которого равна их плавучей плотности (buoyant density), при этом плотность нативной ДНК (r) пропорциональна содержанию пар Г-Ц в пределах 20-80 % Г-Ц: r = 1,66+0,098(Г-Ц).
